Costunolide

In vitro / Animal

Mechanism of Action

Selectively alkylates and inhibits IκB kinase (IKK-β), preventing NF-κB nuclear translocation and downstream pro-inflammatory gene expression; inhibits PI3K/Akt and STAT3 signaling pathways; triggers mitochondrial apoptosis via ROS accumulation and cytochrome c release; anti-angiogenic via VEGF receptor downregulation
NF-kB inhibition; anti-inflammatory via iNOS and COX-2 suppression; anti-tumor via apoptosis induction

Research Notes

Bay LeafWestern

In vitro studies show IC50 of 5–20 µM against multiple cancer cell lines including HeLa, HepG2, and MCF-7. NF-κB inhibition validated across at least 12 independent inflammatory models. Limited in vivo data available; anti-cancer activity in animals is promising but clinical evidence is absent.
