Mechanism of Action

Viridiflorol is a bicyclic sesquiterpenoid alcohol found in Asclepias and several other plant genera. It disrupts bacterial cell membrane integrity through intercalation into phospholipid bilayers, increasing membrane fluidity and reducing the transmembrane proton gradient essential for ATP synthesis. This non-specific membrane disruption produces bactericidal effects against gram-positive respiratory pathogens including Streptococcus pneumoniae and Staphylococcus aureus. Additionally, viridiflorol inhibits biofilm formation by respiratory pathogens, reducing their adherence to bronchial epithelium. Anti-inflammatory activity via inhibition of prostaglandin synthase and reduction of nitric oxide production in activated macrophages has been demonstrated in vitro.

Research Notes

Viridiflorol isolated from Melaleuca quinquenervia and several Artemisia and Asclepias species showed MIC values of 0.5–4 mg/mL against S. aureus, S. pneumoniae, and Candida albicans in broth microdilution assays. Synergistic effects with antibiotics (notably beta-lactams) were documented in one study, with fractional inhibitory concentration indices below 0.5 for S. aureus strains. Anti-inflammatory IC50 for COX-2 inhibition was 35 µM in cell-free enzyme assays. No human studies on viridiflorol specifically are available; contribution to pleurisy root's clinical activity remains theoretical.
