MD-fraction (Maitake D-fraction, purified)

Mechanism of Action

Higher-potency subfraction of D-fraction with enhanced capacity to activate dendritic cells and macrophages. Promotes more efficient antigen presentation and T-cell priming compared to crude D-fraction. Activates the NF-κB and MAPK signaling cascades in immune cells, upregulating co-stimulatory molecules (CD80, CD86) on dendritic cell surfaces.

Masuda et al. (2009, 2013) demonstrated that MD-fraction activated dendritic cells more potently than D-fraction at equivalent doses in vitro. Animal studies show improved anti-tumor efficacy compared to unfractionated D-fraction, with enhanced tumor-specific cytotoxic T-lymphocyte generation. In vitro human peripheral blood mononuclear cell (PBMC) studies confirm dose-dependent enhancement of IL-12 and IFN-γ production. Clinical data limited to small Japanese observational studies.
